Pathogenic for Hereditary breast ovarian cancer syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000017.10:g.(?_41228485)_(41258570_?)del, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the BRCA1 protein in which other variant(s) (p.Cys64Arg) have been determined to be pathogenic (PMID: 8875986, 11462239, 14531499, 21042765, 24249303, 24516540, 27741520). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with BRCA1-related conditions. This variant is a gross deletion of the genomic region encompassing exon(s) 4-13 of the BRCA1 gene. This variant would be expected to be in-frame, preserving the integrity of the reading frame.
