This variant lies in the CDH23 gene (transcript NM_022124.6) at the canonical splice donor site of the intron immediately after coding-DNA position 9319, where G is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: NM_022124.6:c.9319+1G>C. This variant has been classified as likely pathogenic. It is absent from population databases (PM2) and is predicted to result in loss of function in CDH23, a gene in which loss of function is an established disease mechanism (PVS1). It has been reported in trans with other pathogenic CDH23 variants segregating with hearing loss (PM3_supporting). In the present case, it was identified in trans with a likely pathogenic CDH23 variant (PM3). The proband presented with hearing loss and retinitis pigmentosa, consistent with Usher syndrome type I . These findings support the causative role of this variant in hearing loss associated with Usher syndrome type I.
