Likely pathogenic for Retinitis pigmentosa — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001242957.3(MAK):c.101+1G>A, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the MAK gene (transcript NM_001242957.3) at the canonical splice donor site of the intron immediately after coding-DNA position 101, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Variant summary: MAK c.101+1G>A is located in a canonical splice-site and is predicted to affect mRNA splicing resulting in a significantly altered protein due to either exon skipping, shortening, or inclusion of intronic material. Four computational tools predict the variant abolishes a canonical 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 4e-06 in 250778 control chromosomes (gnomAD). To our knowledge, no occurrence of c.101+1G>A in individuals affected with Retinitis Pigmentosa and no experimental evidence demonstrating its impact on protein function have been reported.
