Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001142800.2(EYS):c.9428_9429dup (p.Thr3144fs), citing LabCorp Variant Classification Summary - May 2015: Variant summary: EYS c.9428_9429dupTT (p.Thr3144LeufsX42) causes a frameshift which results in the disruption of the last amino acid and an extension of the protein by 40 additional amino acids. The variant was absent in 102776 control chromosomes. To our knowledge, no occurrence of c.9428_9429dupTT in individuals affected with Retinitis Pigmentosa and no experimental evidence demonstrating its impact on protein function have been reported. Two clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. One laboratory classified the variant as likely pathogenic, and one laboratory classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as VUS.
