NM_024675.3:c.(3201+1_3202-1)_(3350+1_3351-1)dup was classified as Pathogenic for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Autosomal Dominant and X-Linked criteria (2/2020): The EX12dup gross duplication spans coding exon 12 of the PALB2 gene; however, the exact breakpoints of the duplication were not determined. This duplication has been confirmed in tandem in at least one proband and is predicted to result in a translational frameshift with an alternate stop codon (Ambry internal data). This alteration is expected to result in loss of function by premature protein truncation or a transcript that is subject to nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
