Likely pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000314.4:c.(164+1_165-1)_(634+1_635-1)dup, citing Ambry Autosomal Dominant and X-Linked criteria (3/2017): The EX3_6dup gross duplication spans coding exons 3 through 6 in the PTEN gene; however, the exact breakpoints of the duplication were not determined. Additional analysis identified this duplication in tandem in at least one proband and is predicted to result in a premature truncation causing a translational frameshift with a predicted alternate stop codon (Ambry internal data). Based on the majority of available evidence to date, this variant is likely to be pathogenic.
