Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000465.2:c.(1903+1_1904-1)_(2001+1_2002-1)dup, citing Ambry Autosomal Dominant and X-Linked criteria (3/2017): The EX10dup gross duplication spans coding exon 10 in the BARD1 gene; however, the exact breakpoints of the duplication were not determined. This duplication has been confirmed in tandem in at least one proband and is predicted to result in a translational frameshift with an alternate stop codon (Ambry internal data). This alteration is expected to result in loss of function by premature protein truncation or a transcript that is subject to nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
