Uncertain significance for Abnormal blistering of the skin; Oral mucosal blisters; Pretibial dystrophic epidermolysis bullosa; Recessive dystrophic epidermolysis bullosa — the classification assigned by Neuberg Centre For Genomic Medicine, NCGM to NM_000094.4(COL7A1):c.682+5G>A, citing ACMG Guidelines, 2015: The splice region variant c.682+5G>A in COL7A1 (NM_000094.4) has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.682+5G>A variant is observed in 1/1,13,698 (0.0009%) alleles from individuals of European (Non-Finnish) background in gnomAD Exomes and is novel (not in any individuals) in 1000 Genomes. The c.682+5G>A variant is predicted to disrupt splicing by all splice site algorithms. The nucleotide c.682+5G>A in COL7A1 is predicted conserved by GERP++ and PhyloP across 100 vertebrates. For these reasons, this variant has been classified as Uncertain Significance.
