Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 2.4e-05 in 1612982 control chromosomes, predominantly at a frequency of 0.001 within the ASJ subpopulation in the gnomAD database. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.326G>A has been reported in trans with a benign variant in the literature in at least one individual within a cohort of deaf probands who was been subsequently cited by others (example, Bazazzadegan_2012, Koohiyan_2019). These report(s) do not provide unequivocal conclusions about association of the variant with GJB2-related conditions. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
