This variant lies in the PDLIM3 gene (transcript NM_014476.6) at the canonical splice donor site of the intron immediately after coding-DNA position 93 through 4 bases into the intron immediately after coding-DNA position 93, deleting this region. Submitter rationale: This sequence change affects a donor splice site in intron 1 of the PDLIM3 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in PDLIM3 cause disease. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with PDLIM3-related conditions. ClinVar contains an entry for this variant (Variation ID: 1064403). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site.
