Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_017617.5(NOTCH1):c.5455G>C (p.Glu1819Gln), citing Ambry Variant Classification Scheme 2023: The p.E1819Q variant (also known as c.5455G>C), located in coding exon 29 of the NOTCH1 gene, results from a G to C substitution at nucleotide position 5455. The glutamic acid at codon 1819 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
