This variant lies in the IGHMBP2 gene (transcript NM_002180.3) at coding-DNA position 2425, where C is replaced by T; at the protein level this means replaces proline at residue 809 with serine — a missense variant. Submitter rationale: This sequence change replaces proline with serine at codon 809 of the IGHMBP2 protein (p.Pro809Ser). The proline residue is weakly conserved and there is a moderate physicochemical difference between proline and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with IGHMBP2-related conditions.
