Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_005751.5(AKAP9):c.10818G>T (p.Lys3606Asn), citing Ambry Variant Classification Scheme 2023: The p.K3606N variant (also known as c.10818G>T), located in coding exon 44 of the AKAP9 gene, results from a G to T substitution at nucleotide position 10818. The lysine at codon 3606 is replaced by asparagine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
