NM_003280.3(TNNC1):c.341T>G (p.Leu114Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L114R variant (also known as c.341T>G), located in coding exon 5 of the TNNC1 gene, results from a T to G substitution at nucleotide position 341. The leucine at codon 114 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
