Uncertain significance for Spondyloepimetaphyseal dysplasia, PAPSS2 type — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001015880.2(PAPSS2):c.1786C>T (p.Pro596Ser), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15"). This sequence change replaces proline with serine at codon 596 of the PAPSS2 protein (p.Pro596Ser). The proline residue is highly conserved and there is a moderate physicochemical difference between proline and serine. This variant is present in population databases (rs373121598, ExAC 0.004%). This variant has not been reported in the literature in individuals affected with PAPSS2-related conditions. ClinVar contains an entry for this variant (Variation ID: 1062853). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
