Uncertain significance for Combined malonic and methylmalonic acidemia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001243279.3(ACSF3):c.782G>C (p.Gly261Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine with alanine at codon 261 of the ACSF3 protein (p.Gly261Ala). The glycine residue is highly conserved and there is a small physicochemical difference between glycine and alanine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with ACSF3-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt ACSF3 protein function.
