Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001148.6(ANK2):c.9856G>A (p.Glu3286Lys), citing Ambry Variant Classification Scheme 2023: The p.E3286K variant (also known as c.9856G>A), located in coding exon 38 of the ANK2 gene, results from a G to A substitution at nucleotide position 9856. This exon is expressed solely in brain (Mohler PJ et al. Circulation. 2007;115(4):432-41). The glutamic acid at codon 3286 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
