Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_017617.5(NOTCH1):c.7372C>A (p.Pro2458Thr), citing Ambry Variant Classification Scheme 2023: The p.P2458T variant (also known as c.7372C>A), located in coding exon 34 of the NOTCH1 gene, results from a C to A substitution at nucleotide position 7372. The proline at codon 2458 is replaced by threonine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
