Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001985.3(ETFB):c.134C>T (p.Ala45Val), citing LabCorp Variant Classification Summary - May 2015: Variant summary: ETFB c.134C>T (p.Ala45Val) results in a non-conservative amino acid change located in the Electron transfer flavoprotein, alpha/beta-subunit, N-terminal domain (IPR014730) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2e-05 in 251346 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, c.134C>T has not been reported in the literature in individuals affected with Glutaric Aciduria, Type 2b and no experimental evidence demonstrating an impact on protein function has been reported.
