Uncertain significance for Hypertrophic cardiomyopathy 12; Dilated cardiomyopathy 1M — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003476.5(CSRP3):c.358T>C (p.Cys120Arg), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the CSRP3 gene (transcript NM_003476.5) at coding-DNA position 358, where T is replaced by C; at the protein level this means replaces cysteine at residue 120 with arginine — a missense variant. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with CSRP3-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces cysteine with arginine at codon 120 of the CSRP3 protein (p.Cys120Arg). The cysteine residue is highly conserved and there is a large physicochemical difference between cysteine and arginine.
