This variant lies in the LAMA2 gene (transcript NM_000426.4) at coding-DNA position 666, where A is replaced by C; at the protein level this means replaces arginine at residue 222 with serine — a missense variant. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on LAMA2 protein function. ClinVar contains an entry for this variant (Variation ID: 1062477). This missense change has been observed in individual(s) with clinical features of LAMA2-related conditions (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces arginine, which is basic and polar, with serine, which is neutral and polar, at codon 222 of the LAMA2 protein (p.Arg222Ser).
