Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.5007G>C (p.Glu1669Asp), citing Ambry Variant Classification Scheme 2023: The p.E1669D variant (also known as c.5007G>C), located in coding exon 33 of the ATM gene, results from a G to C substitution at nucleotide position 5007. The glutamic acid at codon 1669 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
