Uncertain significance for Brugada syndrome 8 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005477.3(HCN4):c.2062A>G (p.Asn688Asp), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has not been reported in the literature in individuals with HCN4-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces asparagine with aspartic acid at codon 688 of the HCN4 protein (p.Asn688Asp). The asparagine residue is highly conserved and there is a small physicochemical difference between asparagine and aspartic acid.
