This variant lies in the PTEN gene (transcript NM_000314.8) at coding-DNA position 487, where A is replaced by C; at the protein level this means replaces lysine at residue 163 with glutamine — a missense variant. Submitter rationale: The p.K163Q variant (also known as c.487A>C), located in coding exon 5 of the PTEN gene, results from an A to C substitution at nucleotide position 487. The lysine at codon 163 is replaced by glutamine, an amino acid with similar properties. In a massively parallel functional assay using a humanized yeast model, lipid phosphatase activity for this variant was functionally neutral (Mighell TL et al. Am J Hum Genet, 2018 May;102:943-955).
