Uncertain significance for Nephronophthisis 15 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014956.5(CEP164):c.625C>G (p.Leu209Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine with valine at codon 209 of the CEP164 protein (p.Leu209Val). The leucine residue is highly conserved and there is a small physicochemical difference between leucine and valine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with CEP164-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The valine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function.
