Uncertain significance for TWIST1-related craniosynostosis — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006494.4(ERF):c.830A>G (p.Tyr277Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine with cysteine at codon 277 of the ERF protein (p.Tyr277Cys). The tyrosine residue is highly conserved and there is a large physicochemical difference between tyrosine and cysteine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has been observed in one or more individuals who were not affected with ERF-related conditions (Invitae). This variant is present in population databases (rs753014847, ExAC 0.005%).
