NM_007294.4(BRCA1):c.1198G>A (p.Asp400Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D400N variant (also known as c.1198G>A), located in coding exon 9 of the BRCA1 gene, results from a G to A substitution at nucleotide position 1198. The aspartic acid at codon 400 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
