Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_024514.5(CYP2R1):c.107C>T (p.Pro36Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 36 of the CYP2R1 protein (p.Pro36Leu). This variant is present in population databases (rs202122669, gnomAD 0.1%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with CYP2R1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1059990). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
