Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000368.5(TSC1):c.118A>G (p.Met40Val), citing Ambry Variant Classification Scheme 2023: The p.M40V variant (also known as c.118A>G), located in coding exon 2 of the TSC1 gene, results from an A to G substitution at nucleotide position 118. The methionine at codon 40 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
