Uncertain significance for Inosine triphosphatase deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_033453.4(ITPA):c.343C>T (p.Leu115Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces leucine with phenylalanine at codon 115 of the ITPA protein (p.Leu115Phe). The leucine residue is moderately conserved and there is a small physicochemical difference between leucine and phenylalanine. This variant is present in population databases (rs538462304, ExAC 0.002%). This variant has not been reported in the literature in individuals affected with ITPA-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
