Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.2575G>A (p.Glu859Lys), citing Ambry Variant Classification Scheme 2023: The p.E859K variant (also known as c.2575G>A), located in coding exon 15 of the ALK gene, results from a G to A substitution at nucleotide position 2575. The glutamic acid at codon 859 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
