This variant lies in the LRP4 gene (transcript NM_002334.4) at coding-DNA position 4937, where G is replaced by C; at the protein level this means replaces arginine at residue 1646 with proline — a missense variant. Submitter rationale: This sequence change replaces arginine with proline at codon 1646 of the LRP4 protein (p.Arg1646Pro). The arginine residue is moderately conserved and there is a moderate physicochemical difference between arginine and proline. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with LRP4-related conditions.
