Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001183.6(ATP6AP1):c.294C>A (p.Ser98Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with arginine at codon 98 of the ATP6AP1 protein (p.Ser98Arg). The serine residue is highly conserved and there is a moderate physicochemical difference between serine and arginine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with ATP6AP1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
