NM_001023.4(RPS20):c.3+5G>A was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.3+5G>A intronic variant results from a G to A substitution 5 nucleotides after coding exon 1 in the RPS20 gene. This nucleotide position is well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration may weaken the native splice donor site; however, direct evidence is insufficient at this time (Ambry internal data). Based on the available evidence, the clinical significance of this variant remains unclear.
