This variant lies in the RTEL1 gene (transcript NM_001283009.2) at coding-DNA position 2446, where C is replaced by G; at the protein level this means replaces leucine at residue 816 with valine — a missense variant. Submitter rationale: This sequence change replaces leucine with valine at codon 816 of the RTEL1 protein (p.Leu816Val). The leucine residue is moderately conserved and there is a small physicochemical difference between leucine and valine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies.
