This variant lies in the NOD2 gene (transcript NM_001370466.1) at coding-DNA position 2525, where G is replaced by T; at the protein level this means replaces cysteine at residue 842 with phenylalanine — a missense variant. Submitter rationale: This sequence change replaces cysteine with phenylalanine at codon 869 of the NOD2 protein (p.Cys869Phe). The cysteine residue is moderately conserved and there is a large physicochemical difference between cysteine and phenylalanine. This variant is present in population databases (rs749157324, ExAC 0.001%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease.
