Uncertain significance for Desbuquois dysplasia 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_022166.4(XYLT1):c.1588T>G (p.Ser530Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with alanine at codon 530 of the XYLT1 protein (p.Ser530Ala). The serine residue is highly conserved and there is a moderate physicochemical difference between serine and alanine. This variant is present in population databases (rs148992515, ExAC 0.01%). This variant has not been reported in the literature in individuals affected with XYLT1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
