Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_000264.5(PTCH1):c.3276A>G (p.Ile1092Met), citing Sema4 Curation Guidelines. This variant lies in the PTCH1 gene (transcript NM_000264.5) at coding-DNA position 3276, where A is replaced by G; at the protein level this means replaces isoleucine at residue 1092 with methionine — a missense variant. Submitter rationale: The PTCH1 c.3276A>G (p.I1092M) variant has not been reported in the literature to our knowledge. This variant was observed in 1/113754 chromosomes in the Non-Finnish European subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). This variant has been reported in ClinVar (Variation ID: 1057690). Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
