This variant lies in the SCN11A gene (transcript NM_001349253.2) at coding-DNA position 4406, where G is replaced by A; at the protein level this means replaces arginine at residue 1469 with glutamine — a missense variant. Submitter rationale: This sequence change replaces arginine with glutamine at codon 1469 of the SCN11A protein (p.Arg1469Gln). The arginine residue is highly conserved and there is a small physicochemical difference between arginine and glutamine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with SCN11A-related disease.
