This variant lies in the JMJD1C gene (transcript NM_032776.3) at coding-DNA position 1523, where G is replaced by A; at the protein level this means replaces cysteine at residue 508 with tyrosine — a missense variant. Submitter rationale: This variant has not been reported in the literature in individuals with JMJD1C-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces cysteine with tyrosine at codon 508 of the JMJD1C protein (p.Cys508Tyr). The cysteine residue is moderately conserved and there is a large physicochemical difference between cysteine and tyrosine.
