NM_000059.4(BRCA2):c.7758G>T (p.Trp2586Cys) was classified as Uncertain significance for Hereditary breast ovarian cancer syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This missense change has been observed in individual(s) with breast cancer (PMID: 30287823). This variant is not present in population databases (ExAC no frequency). This sequence change replaces tryptophan with cysteine at codon 2586 of the BRCA2 protein (p.Trp2586Cys). The tryptophan residue is highly conserved and there is a large physicochemical difference between tryptophan and cysteine.
