Uncertain significance for Pulmonary fibrosis and/or bone marrow failure, Telomere-related, 4; Dyskeratosis congenita, autosomal recessive 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002582.4(PARN):c.1655A>G (p.Tyr552Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine, which is neutral and polar, with cysteine, which is neutral and slightly polar, at codon 552 of the PARN protein (p.Tyr552Cys). This variant is present in population databases (rs375813873, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with PARN-related conditions. ClinVar contains an entry for this variant (Variation ID: 1056996). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Not Available"; PolyPhen-2: "Benign"; Align-GVGD: "Not Available". The cysteine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
