Uncertain significance for Epidermolysis bullosa simplex 3, localized or generalized intermediate, with BP230 deficiency; Hereditary sensory and autonomic neuropathy type 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001374736.1(DST):c.2539A>G (p.Ser847Gly), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DST gene (transcript NM_001374736.1) at coding-DNA position 2539, where A is replaced by G; at the protein level this means replaces serine at residue 847 with glycine — a missense variant. Submitter rationale: This sequence change replaces serine with glycine at codon 310 of the DST protein (p.Ser310Gly). The serine residue is highly conserved and there is a small physicochemical difference between serine and glycine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C55"). This variant has not been reported in the literature in individuals with DST-related disease. This variant is not present in population databases (ExAC no frequency).
