Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_000135.4(FANCA):c.1940A>G (p.Glu647Gly), citing Ambry Variant Classification Scheme 2023: The p.E647G variant (also known as c.1940A>G), located in coding exon 22 of the FANCA gene, results from an A to G substitution at nucleotide position 1940. The glutamic acid at codon 647 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
