Uncertain significance for Developmental and epileptic encephalopathy 94 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001271.4(CHD2):c.2230A>G (p.Thr744Ala), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with CHD2-related conditions. This sequence change replaces threonine with alanine at codon 744 of the CHD2 protein (p.Thr744Ala). The threonine residue is highly conserved and there is a small physicochemical difference between threonine and alanine. This variant is not present in population databases (ExAC no frequency). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
