Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005228.5(EGFR):c.716G>C (p.Gly239Ala), citing Ambry Variant Classification Scheme 2023: The p.G239A variant (also known as c.716G>C), located in coding exon 6 of the EGFR gene, results from a G to C substitution at nucleotide position 716. The glycine at codon 239 is replaced by alanine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
