NM_001330588.2(TPP2):c.2381T>A (p.Val794Asp) was classified as Uncertain significance for Evans syndrome, immunodeficiency, and premature immunosenescence associated with tripeptidyl-peptidase II deficiency by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine with aspartic acid at codon 794 of the TPP2 protein (p.Val794Asp). The valine residue is moderately conserved and there is a large physicochemical difference between valine and aspartic acid. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with TPP2-related conditions. This variant is not present in population databases (ExAC no frequency).
