Uncertain significance for Arrhythmogenic right ventricular dysplasia 10 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001943.5(DSG2):c.2572G>T (p.Ala858Ser), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DSG2 gene (transcript NM_001943.5) at coding-DNA position 2572, where G is replaced by T; at the protein level this means replaces alanine at residue 858 with serine — a missense variant. Submitter rationale: This variant has not been reported in the literature in individuals with DSG2-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This sequence change replaces alanine with serine at codon 858 of the DSG2 protein (p.Ala858Ser). The alanine residue is weakly conserved and there is a moderate physicochemical difference between alanine and serine. This variant is not present in population databases (ExAC no frequency).
