Uncertain significance for Periodic fever-infantile enterocolitis-autoinflammatory syndrome; Familial cold autoinflammatory syndrome 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001199138.2(NLRC4):c.541C>T (p.Arg181Ter), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change creates a premature translational stop signal (p.Arg181*) in the NLRC4 gene. It is expected to result in an absent or disrupted protein product. This variant is present in population databases (rs759551435, ExAC 0.02%). This variant has not been reported in the literature in individuals with NLRC4-related conditions. The current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in NLRC4 cause disease.
