NM_001122955.4(BSCL2):c.361A>G (p.Met121Val) was classified as Uncertain significance for Charcot-Marie-Tooth disease type 2 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine with valine at codon 57 of the BSCL2 protein (p.Met57Val). The methionine residue is highly conserved and there is a small physicochemical difference between methionine and valine. This variant has not been published in the literature and is not present in population databases. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15").
